與DNA和組蛋白上表觀遺傳修飾類似,RNA上也存在動态可逆的化學修飾(如首個被發現的可逆RNA修飾N6-甲基腺嘌呤),對RNA加工代謝具有重要的調控功能從而影響基因表達,進而調控一系列重要的生命活動過程和疾病的發生發展。這一新興表觀遺傳學研究領域稱之為RNA表觀遺傳學或表觀轉錄組學。RNA表觀遺傳修飾依賴修飾酶、去修飾酶和結合蛋白三類蛋白調控發揮功能。RNA上存在上百種化學修飾,但大多數生物功能未知。
本課題組緻力于開發核酸修飾化學标記測序技術,運用化學生物學、分子生物學、細胞生物學、生物信息學等技術和手段,研究核酸化學修飾和RNA表觀遺傳學在動植物中的生物調控功能。主要的研究方向包括:
- 發展核酸修飾化學标記測序技術,實現核酸修飾的全轉錄組測序和定位;
- 鑒定RNA修飾的修飾酶、去修飾酶和結合蛋白,闡明RNA修飾對植物生長發育、逆境響應等調控機制;
- 改造植物RNA表觀遺傳修飾,實現植物高産等優質農藝性狀;
- 研究RNA表觀遺傳修飾在疾病中的緻病分子機制;
- 發展小分子抑制劑,調控RNA表觀遺傳修飾。
