Lecture Abstract:
Lysine acetylation on histones and other proteins is critical to the regulation of gene expression and other cellular processes. We will describe our focus on the development of designed p300/CBP histone acetyltransferase (HAT) inhibitors as biological tools. We will also discuss aspects of p300/CBP HAT mechanism and regulation gleaned from enzymatic studies. In addition, methods for installing acetyl-Lys mimics and HAT inhibitor elements site-specifically into peptides and proteins will be highlighted.
Lecturer Profile:
Philip Cole 教授目前擔任美國約翰霍普金斯大學醫學院藥理學分子科學和藥理學系系主任,Marshall-Maren 教授、生物活性分子中心主任以及悉尼金梅爾綜合癌症中心化學治療聯合主任。作為表觀遺傳學和化學生物學領域的頂尖科學家,他長期從事蛋白質翻譯後修飾的酶的機制和抑制研究。多年緻力于發展一系列研究轉錄後修飾的化學生物學方法,如目前獲得廣泛應用的蛋白質工程方法“Expressed protein ligation”等,這些化學生物學方法極大地促進了表觀遺傳學研和蛋白質科學研究。他設計的GOAT、蛋白激酶、組蛋白乙酰基轉移酶和組蛋白去甲基化酶特異抑制劑對化學生物學、蛋白激酶與信号傳導、免疫學與代謝治療等作出了重要貢獻。 近年來,他在Science(3)、Cell(2)、Nature(4)、Nature 子刊(11)、Mol Cell(5)、PNAS(11)、JACS(15)等國際頂尖學術刊物發表了160 餘篇高質量研究論文。基于其對化學生物學領域的傑出貢獻,自80年代中期以來,Cole 教授榮獲了一系列國際科技大獎如Irving A Hansen Memorial Foundation Award 和Ellison Medical Foundation New Scholar, 并曾經被評為Innovator of the Year,并擔任多所國際著名大學和研究機構的lecture。
